when we tested TAMC of an API sample it shows one result in first day after five days when we tested the same sample its microbial load decreases up to 50%.
Can anybody explain why that happens?

In general there are no possibilities for decreasing the microbial load once it gets contaminated.
For your concern, it seems there is an analytical error during the earlier testing or recent testing…
Analytical error means improper sample handling/ contaminated glassware usage/ area contamination etc …

We observed TAMC results for spray‑dried API batches.
This doesn’t appear to be an analytical error, results were validated with a third party.
Instead, the reduction may be due to spray‑drying conditions (desiccation stress or heat exposure).
