I just want to ask about microorganisms maintenance,what if i use the mother culture in sub culturing directly to the fresh slants before test ,why should i make subculture from the mother slant monthly??

Subculturing from mother slants or glycerol stocks is essential tomaintain or to demonstrate the some phenotypic or biochemical characters of organisms.
If serial subculturing is performed then due to spontaneous mutation phenomenon some characters may not be shown by the culture…
To avoid this scenario subculturing from mother slants or glycerol stocks is suggested…
USP also suggest subculturing upto 4 passages .

I want to explain the shelf life for each generation and if the mother culture is expired (does this mean all the other generations will be expired)?
