Autotitration assays are the volumetric analysis based on the principle of potentiometric methods. While HPLC methods are chromatographic methods based on the principle of chromatography having staionaery phase and mobile phase. The analyte(s) to be determined are dissolved in the mobile phase and passed over chromatographic columns impregnated with the staionary phase at standardised conditions of flow rate and pressure. Depending upon polarity or Non-polarity characteristics of the analyte compounds and the stationary phase (like, C18 / C8 Silica), the analytes are quantitatively separated from the mixture (Test solution) and hold on the stationary phase and then get eluted quantitatively again in the mobile phase.

There are methods to determine the potency of API by different techniques. In API generally potentiometric titrations are still prefered as in this technique works on following
Potentiometric Titration Potentiometry Principle. When the pair of electrodes are placed in the sample solution or analyte it shows the potential difference between two electrodes by addition of the titrant or by the change in the concentration of ions. Each compound has its unique potential so this technique is used also this is faster and precise method when validated properly.
HPLC is given as additional method in some cases as this becomes a stability indicating method.
