My question is why we use SCDM 10% Glycerol for culture work
Scdm with 10 %glycerol… totally new combination… I have never heard about this…
1.0% glycerol protracts the media from cracking.
Not 10% we use SCDA plates with 1% or 0.5 % glycerol for settle plate exposure
Yes. 1percent glycerol to avoid dehydration. Mainly for settle plate. and not that to exposed plate. It should be added while preparing media before sterilization.

for storage of culture at defrezzer for max. time.
In standard culture maintenance ,we receive 1st passage culture(quick stick) ,after reconstitution (as per vendor instruction ) we streak this stick on SCDA slant(W1), Now question is
Culture on slant(W1) is consider as of 1st passage or 2nd passage ??
Guide me if possible give reference also.
